OPTIMIZATION OF THE TRICHODERMA HARZIANUM CuZn SUPEROXIDE DISMUTASE GENE EXPRESSION IN E. COLI

چکیده
A study of the optimization of CuZn superoxide dismutase gene expression in E. coli L21/pET28-SOD was undertaken. The SOD gene was directionally cloned into expression vector pET28.The recombinant plasmid was transformed into E. coli BL21 and induced by IPTG to express SOD protein. The four effective factors, including the initial concentration of BL21/pET28-SOD, concentration of IPTG, incubation temperature and induction time, were optimized during the induction of target protein. To detect the expression efficiency under different induction conditions, the total protein in the samples were subjected to SDS-PAGE. The SOD protein was successfully expressed in E. coli BL21/pET28-SOD. SOD expression was optimal at the following that E. coli BL21/pET28-SOD at OD600nm =0.6, IPTG 0.125 mmol/L, incubation at 37°C and incubation for four hours.

Enhancement of Solubility and Specific Activity of a Cu/Zn Superoxide Dismutase by Co-expression with a Copper Chaperone in Escherichia coli
Background: Human Cu/Zn superoxide dismutase (hSOD1) is an antioxidant enzyme with potential as a therapeutic agent. However, heterologous expression of hSOD1 has remained an issue due to Cu2+ insufficiency at protein active site, leading to low solubility and enzymatic activity. Objectives:The effect of co-expressed human copper chaperone (hCCS) to enhance the solubility and enzymatic act...

BACKGROUND CuZn-Superoxide dismutase (SOD) is a unique enzyme, which can catalyzes the dismutation of inevitable metabolic product i.e.; superoxide anion into molecular oxygen and hydrogen peroxide. The enzyme has gained wide interest in pharmaceutical industries due to its highly acclaimed antioxidative properties. The recombinant expression of this protein in its enzymatically active and stab...

متن کاملHuman copper-zinc superoxide dismutase complements superoxide dismutase-deficient Escherichia coli mutants.
An Escherichia coli double mutant, sodAsodB, that is deficient in both bacterial superoxide dismutases (Mn superoxide dismutase and iron superoxide dismutase) is unable to grow on minimal medium in the presence of oxygen and exhibits increased sensitivity to paraquat and hydrogen peroxide. Expression of the evolutionarily unrelated eukaryotic CuZn superoxide dismutase in the sodAsodB E. coli mu...
متن کاملBiological Role of Trichoderma harzianum-Derived Platelet-Activating Factor Acetylhydrolase (PAF-AH) on Stress Response and Antagonism
We investigated the properties of platelet-activating factor acetylhydrolase (PAF-AH) derived from Trichoderma harzianum. The enzyme, comprised of 572 amino acids, shares high homology with PAF-AH proteins from T. koningii and other microbial species. The optimum enzymatic activity of PAF-AH occurred at pH 6 in the absence of Ca2+ and it localized in the cytoplasm, and we observed the upregulat...
